Little is yet known about the transmembrane regions (although some are related to the K+ ion channel described in Chapter 12), but we will illustrate extracellular receptors with the example of human growth hormone receptor and the intracellular response systems with p21 ras, transducin (a G protein), and the V-src Tyr kinase. *{Kinemage 1}* shows the structure of the c-ras p21 proto-oncogene (homologous to G proteins), a regulator of cell multiplication. Many tumor cells contain mutant ras proteins which cannot hydrolyze GTP even in the presence of the GAP helper protein, so they remain switched on and lead to cell proliferation. The start-up view matches Fig. 13.4, with the beta strands and helices of the alpha/beta fold color-coded, and the six beta-alpha loops (G1-G6) on the active-site end of the sheet in pink. All of those loops but G5 are involved in nucleotide binding, but the P-loop (or G1) makes especially important backbone-phosphate interactions and includes the two Gly residues most often mutated in oncogenic ras. For comparison, View2 has the same orientation used for the similar transducin structure of Kin.2. Choose View3 and turn on "interact." to see a close-up of the binding site of the GTP analogue GPPNP. Note that one H-bond is lost (from Gly 13 NH) because of the substitution of N for O in the analogue, but the binding is still very tight and specific. Ras, like G proteins and other GTPases, uses a Mg++ ion that bridges between protein and water oxygens and the beta and gamma phosphates of the GTP. *{Kinemage 2}* animates between the two "switch" forms of the alpha subunit of transducin, a G protein involved in the transduction of light signals in the retina. The start-up view matches Fig. 13.6 and 13.7, colored as in 13.7 with the "switch I", "switch II", and switch III" loops green in the GTP and blue in the GDP forms. Animate to see that each of those loops changes conformation significantly. Choose View2 and animate, to see how the beta-sheet H-bonding trades off between different strand pairs in the two forms. Choose View3 and turn on "side ch" for a close-up of the nucleotide binding site; animate to see how the 3 switch loops close in around the gamma phosphate. These changes affect the surface of the subunit quite extensively, in the region that binds to the transducin beta subunit. Choose View4 for an overview and animate; note the extensive changes at the front surface and that the first ordered residue at the N-terminus is 27 for the free alpha subunit. *{Kinemage 3}* shows the 7-fold beta-propeller structure of the transducin beta subunit, with each blade of the propeller in a different color. Animate, to cycle successively through the 7 blades, and then turn them all on to see the full structure. Note that blade 7 has one strand from the beginning of the sequence and 3 from the end, so that the sequence repeat (which we will follow in the next kinemage) is offset from the propeller-blade repeat, as shown schematically in Fig. 13.13b. *{Kinemage 4}* superimposes three of the transducin beta WD repeats, which occur in the sequence as DxxxxxW. Side chains and H-bonds are shown for the conserved His-Ser/Thr-Trp and the Asp-to-backbone interactions that form part of each propeller-blade core and help link the beta-sheet of each successive blade to the next. View1 matches Fig. 13.14a, and View2 is a close-up of the conserved side chains. *{Kinemage 5}* shows the G protein heterotrimer of transducin alpha, beta, and gamma subunits. The 7-fold beta-propeller of the beta subunit is in orange, viewed end-on, and the alpha subunit is blue, with the N-terminal helix and the switch I and switch II regions in lighter blue. Choose View2 to see the interface. This subunit association locks the switch regions into their inactive conformation in which GDP (pink) but not GTP can bind. Conversely, if GTP binds, then the alpha subunit will dissociate from beta-gamma. *{Kinemage 6}* shows the 1:2 complex of human growth hormone (HGH; in pink) with its receptor (blues and greens). The growth hormone is a 4-helix bundle with an unusual up-up-down-down topology (see Chapter 3), while the receptor molecules each have two beta-sandwich domains with immunoglobulin folds (Chapter 15). The two binding sites use the same parts of the HGH receptor molecules, but two different regions on HGH, and they are of quite different binding strength. Choose View2 and turn on "side ch" for a close-up of the tighter-binding region on HGH helices A and D; this is the interaction that forms first and can be stable as a 1:1 complex. Choose View3 to see a similar view of the less extensive contact that uses HGH helices A and C; its formation is facilitated by contact of the C-terminal domains of the two receptor molecules. In c17Engin.kin we will see an engineered analog of this system that uses a small peptide to mimic the hormone. *{Kinemage 7}* shows the final 4 domains of the Hck tyrosine kinase: a beta-barrel SH3 domain in greentint, an SH2 domain in bluetint, and the 2 kinase domains in yellowtint. The central linker (orange) is in polyproline II conformation and binds in the peptide site of the SH3 with the two Pro side chains pointing down (see View2, like Fig. 13.28a, or View5, like Fig. 13.29). Tyr 528 of the C-terminal tail (pink) is phosphorylated, so it binds in the peptide site of the SH2 (see View3, which is like Fig. 13.26). View4 shows the Tyr kinase itself, which in this complex is held open in an inactive form; it is very similar to the CDK2 kinase described in Chapter 6. Copyright 1999 Jane S. and David C.
