RuBisCO must be in its activated form in order to carry out these functions. Activation involves the carbamylation of a conserved lysine (Lys201 in tobacco) by the activator, carbon dioxide and the binding of Mg2+. Unactivated RuBisCO, the focus of this structural study, has a single catalytic function, namely, the decarboxylation of the transition state intermediate, 2-carboxy-keto-D-arabinitol 1,5-bisphosphate. In higher plants, active RuBisCO consists of eight large and eight small subunits. The active site is formed between the interface of the C-terminal beta/alpha-barrel domain of a large subunit and the N-terminal domain of a dimer-related large subunit. Kinemage 1 is an animation of the Calpha trace of unactivated RuBisCO (Nicotiana tobaccum var. Turkish samsun), with and without the transition state analog, 2-carboxy-D-arabinitol 1,5-bisphosphate (CABP) bound in the active site. One large subunit and one small subunit (L1S1) are shown. The structural model was generated from x-ray diffraction data to 2.7 Angstroms. One can observe only slight alterations in the unactivated protein upon binding CABP; high temperature factors in both forms indicate that loop 6 (residues 330-339) does not undergo a conformational change from open (disordered) to closed (ordered), as happens with the activated form. Kinemage 2 is an animation of the active site region of RuBusCO which binds CABP. The two phosphate groups of the transition state analog occupy approximately the same positions of the free sulfate or phosphate (indistinguishable by x-ray diffraction) in the analog-free unactivated enzyme. Phosphate P1 interacts with Arg295, His298, and Ser379, while phosphate P2 binds Lys175 and backbone hydrogens of Gly380, 381, 403, and 404. Kinemage 3 is an animation of CABP in unactivated and activated RuBisCO, demonstrating the reverse orientation of the analog in these two forms (positions of P1 and P2 phosphate groups of CABP are exchanged between the unactivated and activated forms).
